      卵母细胞在冷冻保护剂加载过程中受到的渗透损伤和毒性损伤会影响细胞的存活率,针对这一情况,本文希望通过软件模拟来了解卵母细胞在不同冷冻保护剂加载情况下的渗透行为,以寻求合适的加载方案使得卵母细胞所受的损伤最小。首先利用MLAB编程,分别模拟出卵母细胞在四步法、一般线性法、凸型线性法以及凹型线性法加载冷冻保护剂的方案下的渗透行为;然后,综合积累性渗透损伤AOD值、积累性毒性损伤J值、归一化体积V三项参数来评估卵母细胞受到的损伤。结果表明,当胞外冷冻保护剂为乙二醇时,采用凹型线性加载法加载冷冻保护剂对卵母细胞造成的损伤最小。
英文摘要:
      Considering there will be a reduction of oocyte viability caused by osmotic damage and chemical toxicity during oocyte vitrification cryopreservation. In order to find the best adding pattern of cyroprotectant which can do the least harm to oocyte, this paper makes a process modeling for the osmotic behavior of oocyte when adding cryoprotectant of different concentrations, and also evaluate the damage of oocyte. First, with the help of MLAB, this article makes a process modeling for the osmotic behavior of oocyte when adding cryoprotectant of different concentrations, including four-step, linear variation , convax type linear variation and concave type linear variation. Second, employ three parameters to evaluate the damage of oocytes, including accumulated osmotic damage, accumulated toxicity damage and normalized volume. And we conclude that it’s less harmful to oocytes when adding EG when it’s concentration changes like a concave type linear variation.
